This already brings the death toll for the weekend to seven.
And it follows appeals from Taoiseach Bertie Ahern and a major Garda offensive to cut the carnage.
Last night's crash between a car and an articulated lorry happened shortly before 10.45pm between Ballinhassig and Bandon in west Cork.
Such was the force of the tragic collision that the car - a Polish-registered Opel Astra - was torn apart and it is believed two of the victims died almost instantly.
Two other men, also believed to be passengers in the car, suffered multiple critical injuries but died a short time later before they could be transferred to hospital.
It is understood that several of the dead men may be non-nationals.
The lorry driver is understood to have been treated at the scene for multiple injuries and shock. His injuries are not thought to have been life-threatening and he was being treated for shock in hospital overnight.
A rescue worker, who was at the scene, said it was the worst accident he had seen during his 20 years in his job.
"The car literally disintegrated," he said.
Five units of Cork fire brigade also assisted at the accident scene.
The accident ranks as one of the worst road fatalities in Cork in modern times though four people were killed in crashes in North Cork and West Cork over the past decade.
Driving conditions were said to be good at the time though, but because of the Easter Bank Holiday weekend the volume of traffic volumes was high on the road.
The already high-death toll at the start of the weekend will come as a blow to Government and garda chiefs, who are hoping all their campaigns would lead to a lessening of road deaths.
But even before last night's horrific crash, the holiday weekend had got off to a bad start with three more road deaths.

Śmierć pozostawia żyjącym jakąś pustkę. Przychodzi czas żałoby. Każde uczucie żałoby jest ważne i uzasadnione. Trzeba na nie pozwolić, trzeba je przeżyć. Pozostaje pustka w domu, w sercu. Ból i smutek rozstania. Każdy dzień może być moim ostatnim dniem...
